Why Sterility and Endotoxin Need Dedicated Vials
The chain-of-custody reason these assays cannot share material, and how to count vials before shipping.
Why do sterility and endotoxin tests need separate vials?
Sterility asks whether viable organisms are present in a sealed container. The first entry for any other purpose makes the question unanswerable, because later growth cannot be attributed to the original contents rather than to the act of entry. Endotoxin is less sensitive to prior entry but acutely sensitive to handling contamination, so it also runs on a dedicated unit opened under controlled conditions.
Neither restriction is administrative. Both follow from what the assay is measuring.
Counting vials for a submission

Purity, identity, net content, elemental impurities and residual solvents can all share material drawn from a single vial.

Add one for endotoxin
If bacterial endotoxin testing is ordered, it consumes a dedicated unopened vial.

Add one for sterility
If sterility testing is ordered, it consumes another dedicated unopened vial.

Add per-vial assays last
Vial conformity inspection is billed per vial and applies to every unit shipped, including the dedicated ones.

- How many vials do I need for a full peptide test panel?
- Three, for a panel covering purity, identity, net peptide content, bacterial endotoxins and sterility. One vial supplies all the chromatographic and mass-spectrometric work, one is consumed by endotoxin testing and one by sterility testing. Sending fewer means the microbiological assays cannot run, and a laboratory will normally hold the affected tests and contact you rather than substitute material.
